Single-tablet elvitegravir/FTC/TDF/GS-9350 formulation exhibits high virologic suppression in HIV

Scientists at Gilead have presented positive data on the investigational fixed-dose combination of elvitegravir, GS-9350 (cobicistat), tenofovir disoproxil fumarate (TDF) and emtricitabine (FTC) (elvitegravir/FTC/TDF/GS-9350) for the treatment of HIV at the 2010 Conference on Retroviruses and Opportunistic Infections (CROI) held in San Francisco. In an ongoing, prospective, double-blind, double-dummy, active-controlled phase II trial, treatment-naive HIV patients were treated with elvitegravir/FTC/TDF/GS-9350 (n = 48), administered as a single tablet (Quad regimen), or a combination of efavirenz/FTC/TDF (n = 23) for 48 weeks. At week 24, the primary study objective of the proportion of patients with HIV RNA < 50 copies/mL was met by 90 and 83% of patients respectively. Discontinuation rates and the incidence of adverse events (AEs) were similar in the two treatment groups, suggesting that the efficacy of Quad was noninferior to that of EFV/FTC/TDF in this study population.
